elisa試劑盒總需要時間來保溫，是因為ELISA屬固相免疫測定，抗原、抗體的結合只在固相表面上發生。以抗體包被的夾心法為例，加入板孔中的標本，其中的抗原并不是都有均等的和固相抗結合的機會，只有zui貼近孔壁的一層溶液中的抗原直接與抗體接觸。這是一個逐步平衡的過程，因此需經擴散才能達到反應的終點。
在人高爾基體抗體(AGAA)ELISA試劑盒中一般有兩次抗原抗體反應，即加標本和加酶結合物后。抗原抗體反應的完成需要有一定的溫度和時間，這一保溫過程稱為溫育，有人稱之為孵育，在ELISA試劑盒中似不恰當。

保溫的方式除有的ELISA儀器附有特制的電熱塊外，一般均采用水浴，可將ELISA試劑盒板置于水浴箱中，ELISA板底應貼著水面，使溫度迅速平衡。為避免蒸發，板上應加蓋，也可用塑料貼封紙或保鮮膜覆蓋板孔，此時可讓反應板漂浮在水面上。若用保溫箱，ELISA試劑盒應放在濕盒內，濕盒要選用傳熱性良好的材料如金屬等，在盒底墊濕的紗布，zui后將ELISA板放在濕紗布上。濕盒應先放在保溫箱中預溫至規定的溫度，特別是在氣溫較低的時候更應如此。無論是水浴還是濕盒溫育，反應板均不宜疊放，以保證各板的溫度都能迅速平衡。室溫溫育的反應，操作時的室溫應嚴格限制在規定的范圍內，標準室溫溫度是指20～25℃，但具體操作時可根據說明書的要求控制溫育。室溫溫育時，人高爾基體抗體(AGAA)ELISA試劑盒只要平置于操作臺上即可。應注意溫育的溫度和時間應按規定力求準確。為保證這一點，一個人操作時，一次不宜多于兩塊板同時測定。
