本試劑盒應用雙抗體夾心法測定標本中牛防御素β2(DEFβ2)測定試劑盒水平。用純化的抗體包被微孔板，制成固相抗體，往包被單抗的微孔中依次加入，再與HRP標記的結合，形成抗體-抗原-酶標抗體復合物，經過*洗滌后加底物TMB顯色。TMB在HRP酶的催化下轉化成藍色，并在酸的作用下轉化成zui終的黃色。顏色的深淺和樣品中的呈正相關。用酶標儀在450nm波長下測定吸光度（OD值），通過標準曲線計算樣品中的濃度。

加樣在ELISA中一般有3次加樣步聚，即加標本，加酶結合物，加底物。加樣時應將所加物加在LEISA板孔的底部，避免加在孔壁上部，并注意不可濺出，不可產生氣泡。加標本一般用微量加樣器，按規定的量加入板孔中。每次加標本應更換吸嘴，以免發生交叉污染，也可用一次性的定量塑料管加樣。有此測定（如間接法ELISA）需用稀釋的血清，可在試管中按規定的稀釋度稀釋后再加樣。也可在板孔中加入稀釋液，再在其中加入血清標本，然后在微型震蕩器上震蕩1分鐘以保證混和。加酶結合物應用液和底物應用液時可用定量多道加液器，使加液過程迅速完成。
