用法

稱取本品 36.6g,加熱溶解于 1000ml 純化水中,分裝三角瓶,121℃高壓滅菌 15 分鐘備用。

保存條件： 
培養基由于配制的原料不同，使用要求不同，而貯存保管方面也稍有不同。一般培養基在受熱、吸潮后，易被細菌污染或分解變質，因此一般培養基必須防潮、避光、陰涼處保存。對一些需嚴格滅菌的培養基（如組織培養基），較長時間的貯存，必須放在2~6℃的冰箱內。由于液體培養基不易*保管，現在均改制成粉末。 
實驗內容 
1.稱量→溶化→調pH→過濾→分裝→加塞→包扎→滅菌→無菌檢查 
2.干熱滅菌：裝入待滅菌物品→升溫→恒溫→降溫→開箱取物 
3.高壓蒸汽滅菌：加水→裝物品→加蓋→加熱→排冷空氣→加壓→恒壓→降壓回零→排汽→取物→無菌檢查 
4.過濾除菌：組裝滅菌→連接→壓濾→無菌檢查→清洗滅菌 
的步驟：
（一）準確稱量試劑：根據不同的菌類和用途，選擇適宜的培養基，培養基所需試劑必須純凈。
（二）校正pH值：將稱量好的培養基各種成分放入容器內，標記劃線，加熱溶解，補充水分，測定酸堿度，常用pH6.8～8.0的精密試紙或酸度計測定。用1N NaOH和1N HCl調節pH值到適宜范圍內。
（三）過濾：將玻璃漏斗置鐵架上，再用紗布夾棉花或用濾紙放在漏斗中，將上述培養基倒入其中過濾至透明。
（四）分裝：將過濾后的培養基分裝于中試管或三角瓶內（試管內每支裝5mL；三角瓶中裝100～150ml），塞好棉塞用牛皮紙包扎好，準備滅菌。
（五）滅菌：培養基的滅菌，常用高壓蒸汽滅菌法。一般微生物的營養細胞在水中煮沸后即被殺死，但細菌的芽胞有較強的抗熱性，須經高壓蒸汽滅菌才能達到*滅菌的目的。根據蒸汽溫度隨壓力升高而上升的原理，即壓力越大，蒸汽溫度就越高。因此，在同一溫度條件下，采用高壓蒸汽滅菌比干熱滅菌法效果要好。而且在濕熱情況下，菌體吸收水分后，其蛋白質易于凝固變性，因為蒸汽的穿透力強，殺菌效果好。
