【瘦肉精的危害】

“瘦肉精”進(jìn)入動物體內(nèi)后主要分布于肝臟。肌肉中含量較肝臟低很多。人攝入后在體內(nèi)存留時間較長，其不良反應(yīng)主要有：可引起心率加速，特別是原有心律失常的病例更易發(fā)生心臟反應(yīng)，可見心室早搏、ST段與T波幅壓低，還會發(fā)出肌肉震顫，引發(fā)四肢、面頸部骨骼肌震顫，尤其是交感神經(jīng)功能亢進(jìn)的病例更易發(fā)生。此外，還可引起代謝紊亂、血鉀降低，引起心慌、肌肉震顫、頭痛以及臉部潮紅等。對心率失常、高血壓、青光眼、糖尿病、甲狀腺機能亢進(jìn)等疾病的患者有較大危害。

【產(chǎn)品簡介】

本產(chǎn)品為克倫特羅-萊克多巴胺-沙丁胺醇三合一膠體金快速檢測卡，用于定性檢測豬、牛、羊尿液、組織和飼料中的瘦肉精殘留，整個檢測過程只需要3-5分鐘左右。

【檢測限】

克倫特羅3ng/ml（3ppb），萊克多巴胺3ng/ml（3ppb），沙丁胺醇3ng/ml（3ppb）

【產(chǎn)品組成】

克倫特羅-萊克多巴胺-沙丁胺醇三合一膠體金快速檢測卡（40T/盒）

滴管（1個/袋）、干燥劑（1片/袋）

【樣品處理】

用干燥、潔凈的離心管或適當(dāng)容器采集50ml左右尿液。如果不立即檢測，待檢樣本在2-8℃存放，可保存24小時，注意避免腐壞造成失效或污染。出現(xiàn)陽性結(jié)果應(yīng)按法定程序分瓶封裝樣品用于確證法檢測。

【使用步驟】

1、測試前先完整閱讀說明書，使用前將檢測卡和待檢樣本溶液恢復(fù)至室溫4～30℃。

2、從原包裝袋中取出檢測卡，打開后請在一個小時內(nèi)盡快地使用。

3、將檢測卡平放，用滴管吸取待檢樣品溶液，緩慢垂直滴加2-3滴于加樣孔中，加樣后開始計時。

4、結(jié)果應(yīng)在3-5分鐘時讀取，其他時間判讀無效，根據(jù)示意圖判定結(jié)果。

【結(jié)果判斷】

1. 陰性（-）：兩條紫紅色條帶出現(xiàn)。表示樣品中不含有瘦肉精或其濃度低于檢測限。
2. 陽性（+）：檢測T線無顯色，則表示樣品中瘦肉精濃度高于檢測限。

1. 無效：未出現(xiàn)質(zhì)控C線，表明操作過程不正確或檢測卡已失效。

【注意事項】

1、檢測卡請在保質(zhì)期內(nèi)一次性使用；

2、檢測時避免陽光直射和電風(fēng)扇直吹；

3、盡量不要觸摸檢測卡中央的白色膜面；

4、采樣滴管不可混用，以免交叉污染；

5、如果待檢樣本出現(xiàn)沉淀或渾濁物，請離心后再檢測；

6、試驗遇到的任何問題，請與供應(yīng)商。

【儲存及有效期】

原包裝應(yīng)儲存于4～40℃，陰涼避光干燥處，切勿冷凍；有效期24個月。有效期及批號見外包裝。

用途
用于治療喘息型支氣管炎、支氣管哮喘、肺氣腫所致的支氣管痙攣由對羥基苯乙酮經(jīng)氯甲基化、酯化、溴化、胺化后再經(jīng)水解、中和、氫化制得。
在現(xiàn)代豬肉養(yǎng)殖中被用作瘦肉精，來提高生豬的瘦肉產(chǎn)量。2002年始，被列為養(yǎng)殖行業(yè)違禁藥物，不得在畜禽養(yǎng)殖中添加。
在臨床上婦產(chǎn)科傳統(tǒng)用于治療早產(chǎn)，卻一直沒有被記入藥品說明書。
藥理作用
對氣道平滑肌β2受體有較高選擇性，對兒茶氧甲基轉(zhuǎn)換酶和單胺氧化化酶的抗力增加，作用時間延長, 對心臟的興奮作用很小。體外實驗，對氣道平滑肌和心肌作用所需等強度濃度比較，求得藥物的選擇性指數(shù)，以沙丁胺醇zui高為250，特布他林為138，異丙腎上腺上腺上腺素為1.4。通過患者觀察，比較靜脈注射沙丁胺醇與異丙腎上腺上腺上腺素1秒用力呼氣量（FEV1）及心率變化，沙丁胺醇較小劑量每分鐘0.1μg/kg就能明顯增加FEV1，隨劑量增加，方使心率加速，可見選擇性較高，而異丙腎上腺上腺上腺素在增加FEV1劑量時就能加速心率，選擇性較差瘦肉精  。
藥代動力學(xué)
沙丁胺醇因不易被消化道內(nèi)的硫酸酯酶和組織中的兒茶酚氧位甲基轉(zhuǎn)移酶破壞，故沙丁胺醇口服有效，作用持續(xù)時間較長。口服生物利用度為30%，服后15～30min生效，血漿藥物濃度達(dá)峰時間為2～4h，作用持續(xù)6h以上。消除半衰期為2.7～5h。沙丁胺醇8mg控釋藥片經(jīng)健康志愿者多次服用驗證，90%藥物以0.8mg/h的穩(wěn)定速率釋出。藥物恒定釋出，避免了血藥濃度的波動。藥物釋出率不受酸堿度、胃腸蠕動及食物量影響。氣霧吸入時大部分吸入劑量被吞咽，然后由胃腸道吸收。氣霧劑的生物利用度為10%，吸入后1～5min生效，1h達(dá)高峰。沙丁胺醇大部分在腸壁和肝臟代謝，進(jìn)入血液循環(huán)的原形藥物少于20%。沙丁胺醇口服或吸入后，80%在3天內(nèi)由尿液中排出瘦肉精  。
適應(yīng)癥
用于各型支氣管哮喘以及伴有支氣管痙攣的各種支氣管及肺部疾患；產(chǎn)科用于防治早產(chǎn)，并可解除胎盤血管痙攣，增加胎盤血流量。用于竇性心動過緩、竇性停搏、竇房傳導(dǎo)阻滯、房室傳導(dǎo)阻滯等緩慢性心律失常及心力衰竭等瘦肉精。
中毒
沙丁胺醇（喘樂寧、舒喘靈、羥甲叔丁腎上腺素）的支氣管擴(kuò)張作用與異丙基腎上腺素相等，作用持續(xù)時間較長；增加心率作用僅為異丙基腎上腺素的1/10。用于治療支氣管哮喘、喘息性支氣管炎和肺氣腫患者的支氣管痙攣，口服或氣霧吸入給藥。口服的生物利用度約30%，15～30min起效，維持6h以上；氣霧吸入的生物利用度約10%，1～5min起效，維持時間4～6h。
