【檢驗原理】本品采用競爭抑制法和膠體金免疫層析法原理定性檢測尿液中的尼古丁,以金標尼古丁單克隆抗體作為指示標記物,在硝酸纖維素膜上的檢測線處和控制線處分別包被尼古丁-BSA結(jié)合物和羊抗鼠IgG多克隆抗體。檢測時,尿樣在毛細效應(yīng)下層析。如尿樣中的尼古丁濃度低于200ng/mL時,金標抗體不能全部與尼古丁結(jié)合,未結(jié)合的金標抗體在層析過程中與固定在檢測線處的尼古丁-BSA結(jié)合物結(jié)合,從而在檢測區(qū)(T)出現(xiàn)一條紫紅色條帶;如尿樣中尼古丁濃度高于200ng/mL時,金標抗體全部與尼古丁結(jié)合,從而在檢測區(qū)(T)因為競爭反應(yīng)不會與尼古丁-BSA結(jié)合物結(jié)合而不出現(xiàn)紫紅色條帶。無論尿樣中是否存在尼古丁,控制區(qū)(C)都會出現(xiàn)一條紫紅色條帶。控制區(qū)(C)所呈現(xiàn)的紫紅色條帶是判斷是否有足夠的尿樣,層析過程是否正常的標準,同時也作為試劑的內(nèi)控標準。

【儲存條件及有效期】
儲存條件:原包裝應(yīng)儲存于4~30℃避光干燥處,切忌冷凍。
有效期:24個月。
試劑盒應(yīng)在鋁箔袋拆封后1小時內(nèi)盡快使用;建議在周圍溫度高于30℃或高濕度條件下,盡可能做到即開即用。

在進行檢測前必須先完整閱讀使用說明書,使用前將本品和尿樣恢復(fù)至室溫(20℃~30℃)。
尼古丁診斷試劑
D型產(chǎn)氣莢膜梭菌在動物腸道內(nèi)不斷增殖和產(chǎn)生大量ε毒素,高濃度毒素破壞了腸道壁的完整性,使腸道的通透性大大提高,毒素大量進入血液循環(huán)系統(tǒng),甚至損害與生命活動相關(guān)的神經(jīng)元(如腦干),進而導(dǎo)致患病動物休克或死亡。
腸毒血癥的潛伏期較短,少見臨床表現(xiàn)。臨床上依據(jù)動物腸道吸收毒素多少常將該病分為兩種類型:一種是以強烈抽搐為特征,一般在 2-4 h內(nèi)死亡,患該病動物死亡前肌肉抽搐,流涎過多,四肢不停滑動,眼珠翻轉(zhuǎn),磨牙,隨后頭頸明顯抽縮;另一種是以昏迷和嗜睡為主要特征,病程緩慢,早期臨床癥狀表現(xiàn)為流涎,上下頜“咯咯”作響,之后昏迷,角膜反射等。該病的病理變化常限于呼吸系統(tǒng)、消化系統(tǒng)及心血管系統(tǒng)。剖檢常發(fā)現(xiàn)患該病的動物真胃含有未消化的食物,回腸部分呈現(xiàn)急性出血性炎癥變化,重癥病例整個腸段因嚴重充血而呈紅色。
由D型產(chǎn)氣莢膜梭菌所引發(fā)的動物腸毒血癥有明顯的條件性和季節(jié)性,秋季牧草結(jié)籽后或春末夏初青草萌發(fā)是牧區(qū)動物產(chǎn)氣莢膜梭菌病的多發(fā)期。在農(nóng)業(yè)地區(qū),該病常發(fā)生于莊稼收獲季節(jié)或收菜季節(jié),原因是在該季節(jié)動物常食入過量的谷物、菜葉等,造成腸道消化不良,從而進一步導(dǎo)致腸道菌群失調(diào),為腸道內(nèi)有害菌群的大量繁殖提供了可乘之機。
