【預期用途】

尼古丁(Nicotine)是煙草中的主要生物堿，是導致吸煙成癮的物質動因，也是評價人體攝入煙草煙霧的常用指標。但因為尼古丁半衰期短，無法作為標志物檢測，其代謝物可替寧因為半衰期長作為吸煙和戒煙的標志物。

本品采用競爭抑制法和膠體金免疫層析技術，用于快速定性檢測人體唾液中的可替寧，適用于評價煙草煙霧攝入的初步篩查。

【主要組成成份】

• 可替寧檢測試劑盒（40人份）：每人份鋁箔袋單獨包裝。其中試劑盒由金標可替寧單克隆抗體、可替寧-BSA結合物、羊抗兔IgG多克隆抗體、硝酸纖維素膜、聚酯纖維素膜、塑料背襯、塑料模板組成。
• 一次性滅菌唾液取樣棒（40人份）
• 唾液收集器（40人份）
• 使用說明書（1份）

• 【檢驗方法】

• 撕開鋁箔袋，取出試劑，應在1小時內盡快使用。
• 拿出一次性唾液收集器。（步驟1）
• 手持收集器蓋帽使海綿頭含入口中，直至海綿頭吸滿液體（膨脹變軟）。（步驟2）
• 將海綿頭放回收集器并擰緊蓋帽，使所含樣本流入收集器下端。（步驟3）
• 將試劑盒置于干凈平坦的臺面上，同時打開滴樣孔蓋帽。（步驟4）
• 擠壓收集器上端管壁，垂直滴加2滴無空氣泡的唾液（約100ul）于加樣孔（S）中。(步驟5)
• 等待紫紅色條帶的出現，3-5分鐘時直接觀察結果，10分鐘后判定無效。

結核菌素皮膚反應是遲發型細胞超敏反應。它是抗原（結核桿菌或卡介苗）進入機體使機體的免疫T淋巴細胞致敏，并大量分化增殖。當已致敏的機體再次遭受到抗原入侵時，致敏淋巴細胞就會與之結合，引起細菌反應性炎癥。表現在結核菌素注射部位形成硬結甚至發生水泡、壞死。結素試驗陽性表明機體曾經受到結核菌感染或接種過卡介苗。
一、牛型結核分枝桿菌PPD皮內細菌反應試驗
用結核分枝桿菌PPD進行的皮內細菌反應試驗對檢查活畜結核病是很有用的。該試驗用牛型結核分枝桿菌PPD進行。
1、牛的牛型結核分枝桿菌PPD皮內細菌反應試驗
出生后20d的牛即可用本試驗進行檢疫。
（1）操作方法
a) 注射部位及術前處理：將牛只編號后在頸側中部上三分之一處剪毛(或提前一天剃毛)，三個月以內的犢牛，也可在肩腫部進行，直徑約10cm。用卡尺測量術部中央皮皺厚度，作好記錄。注意，術部應無明顯的病變。
b) 注射劑量：不論大小牛只，一律皮內注射1mL(含2000IU)。即將牛型結核分枝桿菌PPD稀釋成每毫升含2萬IU后，皮內注射0.1mL。凍干PPD稀釋后當天用完。
c) 注射方法：先以75％酒精消毒術部，然后皮內注射定量的牛型結核分枝桿菌PPD，注射后局部應出現小皰，如對注射有疑問時，應另選15cm以外的部位或對側重作。
d) 注射次數和觀察反應：皮內注射后經72h判定，仔細觀察局部有無熱痛、腫脹等炎性反應，并以卡尺測量皮皺厚度，作好詳細記錄。對疑似反應牛應立即在另一側以同一批PPD同一劑量進行第二回皮內注射，再經72h觀察反應結果。
對陰性牛和疑似反應牛，于注射后96h和120h再分別觀察一次，以防個別牛出現較晚的遲發型細菌反應。
