人腫瘤壞死因子α【TNF-α】ELISA試劑盒包被抗體或抗原的選擇：將抗體或抗原吸附在固相載體表面時(shí)，要求純度要好,吸附時(shí)一般要求PH在9.0～9.6之間。吸附溫度，時(shí)間及其蛋白量也有一定影響,一般多采用4℃ 18～24小時(shí)。蛋白質(zhì)包被的zui適濃度需進(jìn)行滴定：即用不同的蛋白質(zhì)濃度0.1、1.0和10μg／ml等進(jìn)行包被后，在其它試驗(yàn)條件相同時(shí)，觀察陽(yáng)性標(biāo)本的OD值。選擇OD值z(mì)ui大而蛋白量zui少的濃度。對(duì)于多數(shù)蛋白質(zhì)來(lái)說(shuō)通常為1～10μg／ml。
