大鼠Smad7 檢測試劑盒用純化的抗體包被微孔板，制成固相載體，TMB在過氧化物酶的催化下轉化成藍色，并在酸的作用下轉化成Z終的黃色。顏色的深淺和樣品中的待測物質呈正相關。用酶標儀在450nm波長下測定吸光度（OD值），計算樣品濃度。

完整的ELISA試劑盒包含以下各組分：
（1）已包被抗原或抗體的固相載體（免疫吸附劑）；
（2）酶標記的抗原或抗體（結合物）；
（3）酶的底物；
（4）陰性對照品和陽性對照品（定性測定中），大鼠Smad7 檢測試劑盒參考標準品和控制血清（定量測定中）；
（5）結合物及標本的稀釋液；
（6）洗滌液，在板式ELISA中，常用的稀釋液為含0.05%吐溫20磷酸緩沖鹽水；
（7）酶反應終止液，常用的HRP反應終止液為硫酸，其濃度按加量及比色液的zui終體積而異，在板式ELISA中一般采用2mol/L。

為了獲得更理想的ELISA實驗結果，請您
1、在ELISA實驗過程中，請精確配制標準品及工作液A和B，以避免由于不準確稀釋而造成的濃度誤差；
2、在ELISA實驗過程中，酶標板在溫育時應加膜覆蓋以避免液體蒸發；
3、在大鼠Smad7 檢測試劑盒實驗過程中，檢測液A和檢測液B，以及底物溶液在使用前，應置于37℃溫育30分鐘待用；
4、在ELISA實驗過程中，洗板后應盡快進行下一步操作，避免酶標板長時間處于干燥狀態。
