產(chǎn)品簡單說明：MMP-8(基質(zhì)金屬蛋白酶-8)，為MMPs家族，是依賴鋅和鈣的內(nèi)肽酶，能夠降解細(xì)胞外基質(zhì)的所有成分。它們在許多生理過程中發(fā)揮重要作用，如胚胎發(fā)育、組織成型、復(fù)制、再生等。它們參與很多病理過程如關(guān)節(jié)炎、癌癥、心血管疾病。新合成MMPs的量主要由復(fù)制水平控制。MMPs的活性主要由酶原的活化和其抑制蛋白質(zhì)(TIMPs)所調(diào)節(jié)。

ELISA的比色測定由酶標(biāo)儀進(jìn)行。有的同行可能會認(rèn)為，既然由酶標(biāo)儀進(jìn)行，那么此時(shí)便可以萬事大吉了，其實(shí)不然，因?yàn)楫?dāng)代較為先進(jìn)的酶標(biāo)儀，均有較多的功能，使用不當(dāng)，會得到令人難以理解的大鼠基質(zhì)金屬蛋白酶8 (MMP-8)elisa檢測試劑盒實(shí)驗(yàn)結(jié)果。如使用酶標(biāo)儀比色測定后，有許多的陰性測定孑L的吸光度值為負(fù)數(shù)；或測定假陽性率大大增加等等。這主要是沒有正確地理解和使用酶標(biāo)儀所致。
此外，在加入底物開始顯色反應(yīng)前，是先檢查一下底物溶液的有效性，即可將A和B兩種液各加1滴于清潔的空板孔或eppendorf管中，觀察是否有顯色出現(xiàn)，如有，則說明底物已變質(zhì)。以O(shè)PD為底物，配好后應(yīng)為五色，否則就不能使用。以TMB為底物，整個(gè)顯色反應(yīng)過程無需避光，而以O(shè)PD為底物，則需避光進(jìn)行。關(guān)于TMB和OPD的顯色反應(yīng)特點(diǎn)及注意點(diǎn)可參考前面有關(guān)章節(jié)內(nèi)容。顯色反應(yīng)完成后，加入酸終止反應(yīng)，振蕩混勻后即可進(jìn)行下面的比色測定或肉眼判斷結(jié)果。

在目前的以HRP作為標(biāo)記酶的商品市場ELISA試劑盒中，如以TMB為底物，則提供的底物為A和B兩瓶應(yīng)用液；如以O(shè)PD為底物，則試劑盒提供OPD片劑或粉劑，臨用前配制。一般商品大鼠基質(zhì)金屬蛋白酶8 (MMP-8)elisa檢測試劑盒顯色反應(yīng)條件為37℃或室溫反應(yīng)15～30min。從理論上說，37℃30min才可以使HRP的底物催化反應(yīng)*，盡管在zui初的10min內(nèi)，絕大部分催化反應(yīng)即可完成。因此，為使弱陽性樣本孔能有充分的顯色，建議在37℃下反應(yīng)25～30min后，終止反應(yīng)比色測定。
