進(jìn)口ELISA試劑盒價(jià)格基本原理
①使抗原或抗體結(jié)合到某種固相載體表面，并保持其免疫活性。
②使抗原或抗體與某種酶連接成酶標(biāo)抗原或抗體，這種酶標(biāo)抗原或抗體既保留其免疫活性，又保留酶的活性。在測(cè)定時(shí)，把受檢標(biāo)本（測(cè)定其中的抗體或抗原）和酶標(biāo)抗原或抗體按不同的步驟與固相載體表面的抗原或抗體起反應(yīng)。用洗滌的方法使固相載體上形成的抗原抗體復(fù)合物與其他物質(zhì)分開(kāi)，zui后結(jié)合在固相載體上的酶量與標(biāo)本中受檢物質(zhì)的量成一定的比例。加入酶反應(yīng)的底物后，底物被酶催化變?yōu)橛猩a(chǎn)物，產(chǎn)物的量與標(biāo)本中受檢物質(zhì)的量直接相關(guān)，故可根據(jù)顏色反應(yīng)的深淺來(lái)進(jìn)行定性或定量分析。由于酶的催化頻率很高，故可*地放大反應(yīng)效果，從而使測(cè)定方法達(dá)到很高的敏感度。
ELISA可用于測(cè)定抗原，也可用于測(cè)定抗體。在這種測(cè)定方法中有3種必要的試劑：
①固相的抗原或抗體
②酶標(biāo)記的抗原或抗體
③酶作用的底物。根據(jù)試劑的來(lái)源和標(biāo)本的性狀以及檢測(cè)的具備條件，可設(shè)計(jì)出各種不同類型的檢測(cè)方法。
