酶聯(lián)免疫技術(shù)在科學(xué)實(shí)驗(yàn)的應(yīng)用：
1）酶聯(lián)免疫組織化學(xué)技術(shù)用于組織細(xì)胞上抗原、抗體成分的定位。
2 測(cè)定法用來(lái)檢測(cè)液體中可溶性的抗原、抗體成分。
3）酶聯(lián)免疫技術(shù)中的競(jìng)爭(zhēng)法是測(cè)定小分子抗原的方法。尤其適用于食品衛(wèi)生分析。食品衛(wèi)生分析中所需測(cè)定的物質(zhì)大都是低分子質(zhì)量的，但免疫動(dòng)物產(chǎn)生抗體的抗原其相對(duì)分子質(zhì)量要求必須很大(至少50 000)，因此，首先必須將小分子物質(zhì)結(jié)合上高分子蛋白質(zhì)，才能作免疫之用，而為了能使小分子物質(zhì)結(jié)合上高分子蛋白質(zhì)，則必須在小分子物質(zhì)上導(dǎo)人一個(gè)活性基團(tuán)，同時(shí)直接競(jìng)爭(zhēng)法所需的酶標(biāo)抗原的制備也需首先在小分子物質(zhì)上導(dǎo)人一個(gè)活性基團(tuán)。
由于該化學(xué)修飾反應(yīng)有一定的技術(shù)難度，有關(guān)專家就采用了間接競(jìng)爭(zhēng)法來(lái)改進(jìn)測(cè)定步驟，用酶標(biāo)抗體代替了酶標(biāo)抗原，因?yàn)槊笜?biāo)抗體的制備技術(shù)相對(duì)比較成熟，并有現(xiàn)成的商品，然而這卻使整個(gè)實(shí)驗(yàn)操作過(guò)程不如直接競(jìng)爭(zhēng)法那么簡(jiǎn)便快捷。
競(jìng)爭(zhēng)酶聯(lián)免疫吸附分析法有特異性強(qiáng)、干擾小、樣品的預(yù)處理簡(jiǎn)便、快速、高效的特點(diǎn)，并且對(duì)設(shè)備裝置的投資較少。
定性測(cè)定結(jié)果判定中常用的一些縮寫(xiě)：
(1)S／CO：其中S為sample(樣本)或specimen(標(biāo)本)的簡(jiǎn)寫(xiě)，表示的是標(biāo)本測(cè)定的吸光度值，CO為cut-off值的簡(jiǎn)寫(xiě)。除競(jìng)爭(zhēng)抑制法外，其他ELISA定性測(cè)定模式中，當(dāng)S／CO值大于或等于1時(shí)，標(biāo)本的測(cè)定為陽(yáng)性，小于1時(shí)為陰性。
(2)S／N或P／N：其中S同(1)，N為negative(陰性對(duì)照)的簡(jiǎn)寫(xiě)，P為patient(患者)的簡(jiǎn)寫(xiě)。較早的試劑盒很多都使用S／N或P／N≥2．1為陽(yáng)性判定標(biāo)準(zhǔn)，現(xiàn)仍有一些試劑盒使用這種方式。這種方式與S／CO方式無(wú)根本性區(qū)別，只不過(guò)是前者將陰性對(duì)照(N)的2．1倍視為cut-off值而已。
