本小鼠乙型肝炎e抗原試劑盒，HBeAg 檢測試劑盒采用競爭ELISA等實驗方法。用待測物質抗原包被于酶標板上，實驗時樣品或標準品中的待測物質與包被的待測物質競爭*標記的抗待測物質單抗上的結合位點，游離的成分被洗去。加入辣根過氧化物酶標記的親和素，*與親和素特異性結合而形成免疫復合物，游離的成分被洗去。加入顯色底物（TMB），TMB在辣根過氧化物酶的催化下現藍色，加終止液后變為黃色。用酶標儀在450nm波長處測OD值，待測物質濃度與OD450值之間呈反比，通過繪制標準曲線計算出樣品中待測物質的濃度。

測定小分子量物質常用競爭法，其標準曲線中吸光度與受檢物質的濃度呈負相關。 標準曲線的形狀因試劑盒所用模式的差別而略有不同。
