本試劑盒應用雙抗體夾心法測定標本中人黏膜地址素細胞黏附分子(MAdCAM-1)水平。用純化的抗體包被微孔板，制成固相抗體，往包被單抗的微孔中依次加入，再與HRP標記的結合，形成抗體-抗原-酶標抗體復合物，經過*洗滌后加底物TMB顯色。TMB在HRP酶的催化下轉化成藍色，并在酸的作用下轉化成zui終的黃色。顏色的深淺和樣品中的呈正相關。用酶標儀在450nm波長下測定吸光度（OD值），通過標準曲線計算樣品中的濃度。
試劑的準備按試劑盒說明書的要求準備實驗中需用的試劑。ELISA中用的蒸餾水或去離子水，包括用于洗滌的，應為新鮮的和高質量的。自配的緩沖液應用pH計測量較正。從冰箱中取出的試驗用試劑應待溫度與室溫平衡后使用。試劑盒中本次試驗不需用的部分應及時放回冰箱保存。
